Synthetic biologists have steadily increased their capacity to incorporate non-standard amino acids into proteins, thereby expanding the genetic code of the cell. In the first part of my talk, I will discuss efforts to engineer pyrrolysyl-tRNA synthetase (PylRS) for improved incorporation of the synthetic amino acid Boc-lysine using Phage Assisted Non-Continuous Evolution (PANCE). In the second portion of my talk, I will discuss engineered coordination of iron-sulfur clusters by the 21st naturally occurring amino acid selenocysteine (Sec). Here we show that Sec can replace cysteine residues coordinating [2Fe-2S] or [4Fe-4S] clusters in two separate proteins without loss of enzymatic activity.-Dr. Corwin A. Miller
