The third mechanism of bacteriophage transduction drives genome hypermobility in Staphylococcus aureus

Bacteriophages are viruses that infect and parasitize bacteria. They are the most abundant entities on the planet, and they are capable of transferring DNA from one bacterium to another by a process known as genetic transduction. Here we report that the temperate phages of Staphylococcus aureus engage in a novel mode of phage transduction that is the most powerful and impactful described to date. We found that large metameric spans (several hundred kb) of the S. aureus genome are packaged and horizontally transferred at frequencies that are 3-4 orders of magnitude greater than any known mechanism of transduction. Large regions of the S. aureus genome are transformed into hypermobile platforms for the high frequency transfer of any genetic element (virulence genes or pathogenicity islands) within their boundaries. We propose that these platforms represent a new concept in mobile genetic elements, for they are defined by genomic coordinates rather than by the DNA elements themselves. Furthermore, this mode of gene transfer pairs a plausible and likely mechanism to many long-standing mysteries in microbial genomics, such as the transfer of recombination hot spots, genomic islands, and pathogenicity islands. -Dr. John Chen
