Research

The tristetraprolin (TTP) family, which regulates mRNA decay, comprises three members in humans and four in rodents. The three related proteins are TTP (also named ZFP36, TIS11), ZFP36L1 (TIS11B, BRF1), and ZFP36L2 (TIS11D, BRF2). We have demonstrated that they play functions in macrophage activation and adipogenesis through destabilization of target mRNAs. Zfp36l3 is a rodent-specific gene, only expressed in the placenta and extraembryonic tissues of mice. All TTP family members have a highly conserved (70% identity) tandem zinc finger (TZF) domain, which is responsible for high-affinity binding to AU-rich elements (AREs) of mRNAs. In eukaryotes, mRNA decay is initiated by deadenylation and the activation of two major decay pathways from the 3′ to the 5′ end or from the 5′ to the 3′ end. Mammalian mRNA deadenylation involved two consecutive phases mediated by the PAN2-PAN3 and the CCR4-NOT complexes, respectively. Recent work indicates that TTP can recruit the CCR4/CAF1/NOT deadenylase complex through direct interaction with CNOT1, resulting in decay of ARE-containing reporter mRNAs. TTP family members share TTP-like activities and promote the deadenylation and destruction of ARE-containing target sequences. However, what specificity in the recruitment of CCR4 and CAF1 paralogs for the individual TTP family member and what the mechanism for the differential role in mRNA deadenylation of the CCR4 and CAF1 deadenylases is unclear. Moreover, how the protein phosphorylation of TTP family involved in deadenylases association is to be investigated.
