Description

The goal of ASCEM is to increase the energy of fundamental life science research of Taiwan and Academia Sinica via introducing the cutting-edge cryo-EM instruments, FEI Titan Krios 300 keV and FEI Talos Arctica 200 keV, and to assist analyzing protein structure and cellular ultrastructure for further solutions of bio-technology issues and serious disease problems, such as the COVID structure analysis and the vaccine development in the recent years. ASCEM also equips with Tecnai G2 F20 TWIN and Vitrobot. The transmission electron microscopes (TEM) systematically provide service from basic to advanced levels on the 24/7 basis to academic and industry users. Currently, the best resolution of protein structure analysis obtained from ASCEM can reach to 1.45 Å.

CEM2 - FEI Tecnai G2 F20 TWIN (TEM)

Max. 200k; Gatan 4K X 4K CCD and DE20 high-sensitivity camera. 

Mainly for biological microstructure analysis, preliminary study of cryo-EM or cryo-tomography, sample screening and a low-resolution model building. Also supports other studies, such as liposome and polymer etc.

CEM3 - FEI Talos Arctica 200 keV (TEM)

Falcon 3EC detector; Volta phase plates; multi-purpose. 

Mainly for automated condition screening and publication quality studies of both cryo-EM and cryo-TM, including overflow of the preliminary study samples from CEM2, screening of grids for CEM4, data collection of high quality data for structural determination (up to 4 angstrom or a little better) and publication.

CEM4 - FEI Titan Krios 300 keV (TEM)

Falcon 3EC and K3 detectors; Volta phase plates. 

Mainly for data collection for high-resolution structures. Preliminary sample, with biological significance, checking by CEM1/2/3 is recommended.

FEI Vitrobot (Sample Preparation Instrument)

Type IV& IV+. Instant cool bio-samples to the temperature of liquid nitrogen in less than microseconds, and freeze bio-molecules in very thin amorphous ice. 

Mainly for SPA and cryo-Tomography experiments. Also supports other studies such as liposome and polymer, etc.
